I. PURPOSE

To provide the proper collection protocol for Tzanck Smear testing.

II. MATERIALS

Skin lesion:
• 2 slide smears


III. PROCEDURE

1. Label slides with patient’s name, date of birth and source site on the frosted end in pencil.
2. Cut approximately ¾ of the way around the vesicle in an elliptical or circular pattern. Flip over the flap
    of tissue and scrape the under side of the roof of vesicle with a blade. Gently smear on clean glass slide.
    Do not produce bleeding.
3. Pustules can be broken with a sterile hypodermic needle; if cultures are ordered, a swab may be used
    to break the pustule and then submitted for the culture specimen.
4. Two smears should be obtained directly from the suspect lesion and spray fixed.
5. Place slides in appropriate cardboard/paper holder and secure with tape.
6. Place in a biohazard bag along with the completed requisition in the side pocket of the specimen bag.
